Yes it will have an effect on arrow flight.
If your arrows are fishtailing they are moving side to side and you need to move your rest either right or left. If they are porpousing then you need to move your nock point up or down on your string.
There are a bunch of other things that could also be the problem like arrow spine, tip weight, string strech, etc... But we will assume that you got that OK.
Go to Eastons web site and download their tuning guide. It will run you through the entire tuning process.

Two things you need to immediately check.
You are either #1, torquing the bow very badly or #2, your center shot is way off.
#1, Have someone stand in front of you and draw you bow back with out an arrow and anchor just like you are going to shoot. Have that person look at the bows riser and see if it is in line straight with the string. If no one is available you can come to full draw and peek down at the string and cam. The string and cam should be in line with each other.
#2, rest your bow on the floor on its bottom cam with an arrow nocked. Look down on your arrow. It should be pointing straight from the string out past the riser. If your center shot is off it will likely be pointing out to the left for a right handed shooter.
If you have a place to paper tune you should definetly do that.
