It all depends on what you are talking about. As far as dropping one on the spot every time there really are no guarantees. I have seen several studies that confirm it. Even a head shot doesn't guarantee it will drop on the spot, animals can still run after being decapitated.
I would say the most reliable shot is the high shoulder shot though, breaking both shoulders and shocking the spine is a pretty effective way to immobilize something. Doesn't mean it died that instant, but it certainly isn't going to far.
I feel it has more to do with shot placement and bullet construction than anything else.

I read that therory several years ago and I believe that it makes sense. We all know that there is no way to know when the heart valve is open freeing the path for shock to the brain but it sure makes sense to me. The surest shot as Paul said is the high shoulder shot that hits or sends bullet or bone framents into the the sinal area. It may not be an instant kill but they seldom leave their tracks.
