My favorite deer hunting CALIBER is .308". My favorite deer hunting RIFLE CARTRIDGE is the .30-06, my favorite HANDGUN cartridge is the .44mag.

That said, I can be found hunting on any given day during hunting season with any of about 10 different cartridges... .243win, .243WSSM, .270win, .308win, .30-06, .30-30, .308Patriot, .338-06, .41mag, .44mag, .454cassul, .480Ruger, or .45-70...or any "temporary" rifle or handgun I happen to have at the time...Not to mention the 12ga's, compound bows, and .50cal ML's I hunt with each year.

Variety is the spice of life!!

The answer to this one will sometimes indicate the respondant's age.
My favorite deer cartridge is a .270Win 130gr handload in my Winchester Model 70, made in 1952. I would never have gotten another rifle for deer, but I went after elk and IMO, believe that .270Win is underpowered for elk in other then ideally presented shots.
I would not trade my .270Win for a .280Rem or 7mmWSM for use on deer, but if I did not have the .270Win, I would opt for one of those.

I am going to assume that you ment cartridge and not caliber since you said the good old .30-06.

I have two favorites.....in open areas I shoot my Sig SHR970 chambered for the .280 REM round. If I am in heavy cover I shoot my Marlin 1895 GG in the most awesome .45-70.
