The answer to your question is, yes the shots will almost be different. The reason for this is your first shot is called a "cold bore shot". What we we do in the wold of sniping is log in a book each one of our rounds and where it his on paper. The key to accurate shooting is consistancy. The cold clean bore shoots different because you are shooting from a cold barrel (which changes the point of impact) and you are pushing out all of the chemicals you used to clean and store your rifle. We log all of our cold bore shots and after a while you can look at your log book and see that your first shot will always be for example 1/4" high and 1/4" right.
A couple of ways around this are to leave the barrel dirty after you sight in. This is fine for short periods of time. Another is to use lighter fluid before you go into the woods. This is probably not a good idea with a muzzleloader and reserved more for CF rifles.
Some of the other guys here also were right in saying that the best way to shoot is to run a spit patch through after every shot. If you don't do this the barrel will become increasingly fouled and your point of imact will change slightly with every shot.
I know this is a lot ot deal with just for killing a deer, but that is why you would foul the barrel.
