You do not need to break in the bow but you do need to break in the string. Your string will strech a good bit for about 200 - 300 shots. Go ahead and sight in your bow at 20 yards to make sure that you will stay on target but do not get worked up if you see your point of impact start to wander. Take measurements now (peep location, nock location, tiller measurement, cam timing(2 cam) axle to axle measurement, brace height) so after your string has streched you can put everything back to the way it was. After that you will need to twist up the cable and string to get it back in specs and then you can get good and tuned.
As far as sighting in goes I prefer to shoot at a three or five spot target with one arrow in each spot. This helps save your arrows from abuse. And like was stated above just move your sight in the direction that your arrows are from the bulls eye.
