The .45 ball is slightly oversize to the cylinder. Some lead is shaved from the ball as it's seated in the cylinder to form a tight seal.

The cylinder is also slightly larger than the .44 bore. The ball is swagged into the bore by the forcing cone when fired.

I'm sure the dimensions are slightly different for each manufacturer.
