Hello
I have an old 50 cal. ML that I put together about 20 years ago when I was a teenager. The gun has not been fired for 10 years or so and I want to get it out and use it this year.
First question- Is there anything I should do to the barrel before loading it the first time? It does not have any noticable rust at the muzzle.
Question 2- can I use those new aerobullets with this gun? I was going to use black powder because I like the cool smoke plume and I understand that some of these new fangled bullets need more than a 1-48 twist and like the synthetic powders and so on.....

As long as you know the rifle is in excellent condition as far as structure wise, there is no reason you can not start shooting it.
First thing I would do is clean the barrel with a soap and water bath. It will surprise you what kind of dust can accumulate over many years down a barrel. Then it would not hurt to swab the barrel with some solvent patches too. After that I would run an alcohol patch or two and then some dry patches down the barrel. Pop a could caps to make sure you have fire coming through the fire channel and to clean and clear anything in there, and you should be ready to shoot.
As for shooting Aero Tip bullets you might get good accuracy with them. All you can do it try some of them. If that does not work, then go back to the old standard loads you used to shoot.

If you have a 1/48" twist, you should be able to shoot Power Belts and Maxi-Balls from it - I do, and my twist is a lot slower than yours!
