You would have to fire a LOT of rounds to completely fill the grooves in a rifled barrel. Accuracy can fall off fairly quickly though when shooting rifled slugs in a rifled barrel.
Will it hurt it? No, not if you clean it when you are done. Many people shoot rifled slugs in rifled barrels, as a matter of fact that is what they used before sabots became so popular.
It is much the same thing as shooting a conical bullet out of a muzzle loader or lead bullets out of a pistol or rifle.
